GST-tag purification is based on the specific affinity between Glutathione S-transferase (GST)-tagged recombinant proteins and immobilized glutathione ligands. This interaction enables rapid, selective purification under mild, non-denaturing conditions. Bound proteins are typically eluted using buffers containing reduced glutathione.
